Would suggest you look first at ports board for other who have done this. We checked ours at the Best Western near the express bus for the airport. You can check at thec train station, but am sure other places as well.

We started our cruise in Venice last year and my research showed that there is a left luggage office in the Piazzale Roma. When you leave your cruise ship, there is a people-mover rail link to the square (€1.50 each including bags). This is were you can pick up the bus to the airport as it is the terminus, or a taxi.

 

As we didn't need to leave our bags I never found the left luggage place but I would think Googling it would help.

 

As for the rest of the day... Venice is very walkable. You could probably walk to St Marks square in the same time that a vaporetto would take. At the ends of all the small streets are signs for the three major places you would need - Piazzale Roma to get back, San Marco for St Mark's square, and Rialto for the bridge. Lots of steps over bridges.

You would have plenty of time to wander to both of these and get back in time for your run to the airport. Just note that eating and drinking get more expensive the closer you are to St Marks! and I did read about someone spending €120 on four coffees, so it pays to be a bit cautious.

 

If you need water, the fountains in many of the squares have potable water which is cool even on a hot day (30+C when we were there).

 

Enjoy your day, however short. It is a wonderful city. Drink a little prosecco and eat some cichetti. We can't wait to go back.

We started in Venice and stayed at The Ca' Doge which is located on Piazzale Roma so I saw the left luggage spot used by many. As you exit the People Mover (which is how we moved from our hotel to the port and is great!), turn left and you'll find the luggage storage spot almost immediately on your left. It is a small storefront so there may well be a line, but it seemed to move quickly. Enjoy your time in this magical city!

Kitekat's suggestions sound good. I would also have lunch at one of restaurants with tables right on the Grand Canal. Watching the boats go by was a great way for us to learn more about Venice (a lot of action - such as ambulance and police boats zooming by and great people watching with young couples and families enjoying outings). Having a liter of their wonderful wine with lunch is very relaxing, if you like wine. Just don't miss the plane! ;)

The main train station and has left luggage. If you want to walk from there to St Marks or the Rialto Bridge then look out for signs on the corners of buildings which will direct you, and the same when going back, the Piazzale Roma is sign posted in the same way. Nothing takes too long to walk and it's so beautiful. We did a 5 days escorted tour there in October, it was wonderful.

Some of the cruise terminals in Venice do have a luggage deposit (but not all of them). Mainly the terminal 103.

 

There´s a luggage deposit right to the left of the exit of the People Mover at Piazzale Roma. It´s just a small shop and it fills up quickly. I´ve used it once.

 

There are luggage deposit boxes at St. Lucia train station too. From Piazzale Roma just cross over the bridge (unfortunately it has steps).
