Cassis is charming and you just get there and get on a boat that is going to the Calanques and when it is full it heads out. In your case, I would recommend starting in Marseille, as otherwise you will lose a lot of time travelling over to Cassis (which is a bit tricky to get to by public transport). In either case, you don't need to reserve. Just go down to the Old Port (Marseille) or the harbourfront in Cassis and buy a ticket. Tours leave very regularly throughout the day.
