Not sure you're going to get a lot of response here about long-distance buses because of the demographics of posters here. You might try posting on Lonely Planet's Thorn Tree forum for Bolivia and/or Argentina, which gets a lot more of the backpacker crowd. Argentine buses are supposed to be pretty comfortable. Is it possible you mean from Buenos Aires? My recall is, those bus rides are about 17 hours and the cost is sometimes fairly close to going by plane. If you choose this option, by all means opt for a bus offering Cama Suite service, equal to first class on a plane.
