All trains going to Venice Santa Lucia (the stop that is actually in Venice) stop at Mestre, and MOST, but not all trains going to Mestre go on to Santa Lucia.

 

These are all regional trains; the whistle stop ones take 33 minutes while the faster ones (regionale veloce) take 25 minutes and have fewer stops.

 

Just check the departures board at the station, it will show you the last stop for each train.

When you check in at the hotel they will give you a train schedule and you can buy your tickets at the front desk when you are ready to travel. Be sure to validate your ticket in the machines before you go up to the platform to wait for the train. Someone did come around to check them.
