If I have 2 cruises booked, B2B however 2 separate reservations, will I be allowed to bring 4 bottles of wine initially and have Royal hold 2 bottles for 2nd sailing?

Yes. They may or may not hold the bottles for the 2nd leg. I've had it go both ways.

Just have your SetSail Pass available to show them (if they ask) as "proof" that you're doing a B2B.

But if each person is carrying two bottles, they probably won't even notice.
