We’re going on the Sun in May, the itinerary includes 4 stops in different countries, Italy, Spain, Greece, and Turkey.  Do we have to go thru customs at each port of a different Country?

You will only have to go through customs in Türkiye assuming that's your departure port. You will only go through European customs when you arrive at the airport.

[Arizona Wildcat]

You will go through customs AND immigration on your outbound fight.  Once if flying directly to the EU and a possible immigration check if changing flights in UK.

Then customs and immigration leaving the EU AND in the US

Nothing for port calls.
