We did during.  Simple visit to LA's (Loyalty Ambassador) desk.  If he or she is not there, should be able to find a stack of cards there, that you can fill out and drop in the box at the desk.

Even if you sign up after and you provide the dates and reservation information from previous cruies they will give you credits once you have signed up.

Sign up on board is fine.  As suggested, just see the Loyalty Ambassador on the ship.  They typically have desk hours at a location near the Next Cruise office.  As mentioned, your points are earned on each cruise and accrued for your tier totals after each cruise is completed.  Any change in tier level is recognized on the subsequent cruise from which is was reached.
